OX40 is a T cell co-stimulatory molecule of the TNF receptor superfamily. Its interaction with OX40L plays an important role in the expansion and survival of antigen-specific T cells. OX40 is mainly expressed by CD4+ and CD8+ T cells, and requires the combination of antigen-presenting cells and TCR and the co-stimulation of CD40-CD40 ligand and CD28-B7. OX40-OX40L interactions have been found to be important in the regulation of T-cell tolerance, peripheral T-cell homeostasis, and T-cell-mediated inflammatory diseases, making them attractive candidates for clinical intervention.
