巨噬细胞的清除,是全面了解巨噬细胞在病理条件下功能的一种十分有力的研究方法,氯膦酸盐脂质体(Clodronate Liposomes)是目前最为成熟,方便,经济的一种巨噬细胞清除工具,可以有效的清除动物体内不同组织部位和血液中的巨噬细胞。
氯膦酸二钠(Clodronate)是一种双膦酸盐药物,在临床上主要用于癌症引起的溶骨性骨转移、骨质疏松症和高钙血症。其作用机理是氯膦酸二钠在细胞内代谢成不可水解的ATP类似物,抑制线粒体中的ADP/ATP转运机制,从而阻断线粒体呼吸链诱导细胞凋亡。但是该药物亲脂性低,不能跨细胞转运,在口服或是静脉给予游离药物后,2小时后即可达半衰期,48小时内80%药物随尿液排出。
脂质体是单层或多层脂质双分子层的球形囊泡,其主要成分是磷脂和胆固醇。脂质体本身具有双分子层结构,组成成分无毒性,因此可以作为运输分子物质的载体,它通过与细胞膜结合的方式将分子物质运输到细胞内。
氯膦酸盐包裹进磷脂体的水相中形成氯膦酸盐脂质体,氯膦酸盐不能自由的通过脂质体磷脂体双分子层。注射到生物体内后,氯膦酸盐脂质体会被巨噬细胞吞噬,在巨噬细胞内的溶酶体磷酸酶的作用下,溶解在脂质体内的氯膦酸盐会被逐步释放出来并在细胞内积累,当达到一定浓度时,巨噬细胞将会受到不可逆损伤,诱发细胞凋亡。死亡细胞内的氯膦酸盐会被释放到细胞外,随尿液排出体外。
