DSS诱导斑马鱼溃疡性结肠炎模型的建立

斑马鱼作为肠道损伤模型的应用已得到充分证实,葡聚糖硫酸钠(Dextran Sulfate Sodium Salt,DSS)诱导的斑马鱼IBD模型模拟了哺乳动物IBD表型的不同特征,包括肠道中性粒细胞炎症、过多粘液生成、肠淋巴管生成增加以及前炎性细胞因子上调。

实验动物

3dpf(days post fertilization) 斑马鱼

 

实验材料

DSS(Yeasen 60316ES,MW:36000~50000)

 

验步骤

                                       1. 将斑马鱼胚胎培养在含甲基蓝的E3胚胎培养基中,28.5 ℃,培养至1 dpf;

                                       2. 1dpf 后使用不含亚甲基蓝的E3胚胎培养基,培养至3 dpf;

                                       3. E3培养基配制浓度为10% DSS的储存液;

                                       4. 用培养基稀释DSS至最大非致死剂量(DSS浓度参考:0.5%);

                                       5. 0.5%的DSS处理斑马鱼,从3 dpf处理到6 dpf,观察指标[1]

 

结果分析

                                       造模成功指标:中性粒细胞数量增多、酸性粘液蛋白增多、炎性因子表达量上升。

                                       1.嗜中性粒细胞数量:中性粒细胞数量增多,并且向肠和上皮中移动

                                       2.酸性粘液蛋白:石蜡病理切片结合AB-PAS染色法验证,DSS浸泡后斑马鱼肠道中酸性粘液蛋白增多

                                       3.炎性因子表达量DSS处理后斑马鱼肠道中炎性因子表达量上升[1,2]

DSS处理组相较于对照组,酸性粘蛋白染色和粘液分泌增加,斑马鱼外周血中的嗜中性粒细胞数量增多并且向肠和上皮中移动,炎性因子表达量显著上调,肠道出现炎症反应。
