Nature IF=69.5|哈佛医学院研究发现结肠炎的表观遗传记忆会促进肿瘤生长

慢性炎症被认为是恶性肿瘤发生发展的主要风险因素之一 。长期的炎症微环境通过持续释放炎性介质和活性氧,不仅诱发组织损伤,更会驱动上皮细胞进入反复的“损伤-修复”循环。溃疡性结肠炎(UC)作为一种典型的慢性炎症性肠病(IBD),其病程呈持续性进展,是结直肠癌(CRC)的高危因素 。临床数据显示,UC患者发生 CRC 的概率比普通人群高出2至5倍。即使在炎症得到有效控制后的缓解期,患者的癌症风险依然显著高于常人。这种“炎症消退但风险持续”的现象提示,肠道组织内部可能存在一种长效分子记录机制,即“表观遗传记忆”(Epigenetic Memory)。尽管炎症诱导的DNA损伤累积在致癌中的作用已明确,但组织在炎症消退并实现形态学修复后仍长期维持癌变易感性的“场癌变”现象,其核心分子机制,尤其是非突变性表观遗传重塑的贡献仍是药理学研究中尚未解决的关键科学问题 。
针对这一前沿问题,来自哈佛大学与Broad研究所的 Jason D.Buenrostro 教授团队于2026年3月25日在 Nature 发表了题为 Epigenetic memory of colitis promotes tumour growth 的研究长文。作者利用自主研发的 SHARE-TRACE 技术(一种同时检测单细胞转录组、染色质可及性和克隆可遗传性的新技术),在DSS诱导的结肠炎模型中发现,结肠干细胞在炎症消退后仍保留着长达100天以上的“表观遗传记忆”。该研究证实,这种记忆以激活蛋白1(AP-1)转录因子活性的累积增加为核心特征,并能通过干细胞分裂稳定遗传。这一发现不仅阐明了炎症致癌的新型药理机制,还为IBD患者的长期监测及癌症预防提供了潜在的诊断标志物和原创治疗靶点。



方法
1.DSS结肠炎模型构建
实验选用 C57BL/6 小鼠,分3个疾病阶段:
①急性损伤组:1轮低浓度DSS饮水(2.5%DSS,自由饮用7d),每日记录体重、粪便性状、便血情况,计算DAI疾病活动指数,7天后,全部换回无菌蒸馏水,正常饲养14天,肠道黏膜进入修复阶段;
②慢性复发性结肠炎组:3轮循环造模(7d DSS饮水+14d清水恢复,重复3次),模拟人类反复发作UC,同步每日DAI评分监测;
③炎症恢复期:3轮DSS结束后,持续清水饲养21d~102d,21d时结肠隐窝结构、体重、免疫浸润完全恢复至正常外观,肉眼/病理无炎症,但干细胞留存表观记忆。

2. 炎癌转化复合模型(验证记忆促癌功能)
采用Apc诱导突变小鼠,分组对照:
对照组:无DSS、直接诱导Apc突变;
结肠炎恢复组:先完成3轮慢性DSS结肠炎造模、恢复60天,再诱导Apc致癌突变。结果显示:经历过结肠炎的小鼠,肿瘤体积、数量显著上升约40%,直接证明结肠炎形成的表观遗传记忆可预致敏结肠干细胞,进而加速结肠癌变进程。

3. 体外验证配套模型
分离恢复期小鼠结肠干细胞,构建结肠类器官体外模型:脱离体内炎症微环境后,炎症来源类器官仍持续保留AP-1表观印记,增殖能力显著高于正常小鼠类器官,研究证明记忆是细胞内在可遗传的,且不依赖体内炎症微环境。



在相关研究中,可选用翌圣生物Dextran Sulfate Sodium Salt(DSS) 结肠炎建模用葡聚糖硫酸钠盐MW:36000~50000(60316ES)进行小鼠结肠炎造模:
