DSS诱导猪溃疡性结肠炎模型的建立

背景概述

肠道通透性增加和相关体重减轻是DSS结肠炎动物模型的常⻅特征。D-甘露醇是⼀种惰性小碳水化合物分⼦,可沿整个隐窝-轴吸收,⽤于评估体内肠道通透性。

实验动物

4-5天大约克郡小猪

实验材料

DSS(yeasen DSS 60316ES,MW36000-50000)

实验步骤

1. 以商业代乳粉配⽅饮⾷,每天喂食三次,直到实验结束

2. 将动物分为阳性对照组(Pos),阴性对照组(Neg),实验组(Trp);

3. 阳性对照组(Pos):灌注DSS;阴性对照组(Neg):灌注生理盐水;实验组(Trp):灌注DSS+治疗药物

4. 每只小猪每日摄入量1.25g DSS/kg,灌注5天

1. 肠道通透性分析

DSS诱导致小猪体内D-甘露醇浓度升高,肠道屏障被破坏。

DSS处理的Pos组隐窝结构扭曲、炎症细胞浸润到粘膜和粘膜下层、隐窝脓肿和隐窝炎

DSS处理的Pos组中两种细胞因⼦的浓度均升高[1]
