TRIeasyTM Total RNA Extraction Reagent是一种适用于各种动植物、细菌组织、细胞的总RNA抽提试剂,具有极强的裂解能力,可在短时间内裂解细胞和组织样本,并有效抑制样本中RNA的降解,保持RNA的完整性。样品在该试剂中能够充分裂解,之后加入氯仿离心分层,形成上清层、中间层和有机层(鲜红色下层),RNA分布在上层水相中,收集上清层后,经异丙醇沉淀便可得到总RNA。提取的总RNA纯度高,基本不含蛋白质及基因组DNA,可直接用于Northern、点杂交、mRNA纯化、体外翻译、RT-PCR、poly(A)+选择、RNA酶保护分析以及构建cDNA文库等多种分子生物学实验。
此外,样品中的DNA和蛋白也能以沉淀的方式还原。乙醇沉淀能析出中间层的DNA,而在有机层中加入异丙醇能沉淀出蛋白。本品操作简单快速,所有操作可在一小时内完成。对少量的组织(50-100 mg)和细胞(5×106)以及大量的组织(≥1 g)和细胞(>107)均有较好的裂解效果。

注意事项
1. 本产品中含有苯酚,具有毒性和腐蚀性。如果吸入体内、接触皮肤、吞食等会引起中毒、灼伤及其他身体伤害。使用时应穿戴防护物,如防护服装、手套、眼罩、面罩等。如果不小心接触到眼睛时,应立即用大量的水冲洗并前往医院治疗。
2. 请穿实验服并佩戴一次性手套操作,避免RNase污染。
3. 需自备氯仿、异丙醇(新开封或提取RNA专用)、75%乙醇 (DEPC处理水配制)、DEPC和DEPC处理水。
4. 样品用Total RNA Extraction Reagent匀浆后,如果不加入氯仿进行下游实验,可先-70℃冻存,可保存一个月以上。
5. RNA沉淀在75%乙醇中,4℃可保存1周,-20℃可保存1年。
6. RNA半衰期比较短,易降解,建议抽提后尽快进行后续实验。

Q:10606和19201的区别是什么?
A:19201是浓缩形式,需要的试剂用量更少一点,提取的准确性和提取反应的速度更快一些。一般10606就可以满足常规提取需求,如果客户想好更好的提取效果和rnase抑制效果可以用19201。
Q: 用在贴壁细胞上总是不能完全从培养瓶(皿)脱落,是裂解不完全吗?
A: 这并不意味着裂解不完全。此时,细胞膜实际已完全裂开,并释放RNA,继续后续实验即可。
