Ampicillin,中文名氨苄青霉素,同义名氨苄西林,一种β-内酰胺类抗生素,属于青霉素家族的一员,同样敏感于β-内酰胺酶,后者可裂解氨苄西林的β-内酰胺环。Ampicillin广谱抑制革兰氏阳性菌,阴性菌以及厌氧菌,其抑菌机制在于:干扰青霉素结合蛋白(penicillin binding protein,PBP)活性,PBP参与肽聚糖合成的最后一步,其可催化丙氨酸和赖氨酸残基间形成五甘氨酸交联,此交联维持细胞壁的完整性,并维持细菌的正常生长。另外,Ampicillin仅对生长的大肠杆菌(Escherichia col)具有杀菌作用。

Ampicillin普遍用作细胞培养中的一种选择性抗生素,用于配制含氨苄的LB或LB平板等,也可用于培养细胞的杀菌。Ampicillin抑制革兰氏阴性和革兰氏阳性细菌,最小抑制浓度(MIC)分别为0.03-3 μg/mL和0.02-1.5 mg/mL。高压灭菌的琼脂或培养基需冷却到45-50℃再加入氨苄青霉素溶液,含氨苄青霉素的平板在2-8℃可保存2周。在37°C培养条件下,氨苄青霉素可稳定3天。

本品为氨苄青霉素钠盐,细胞筛选常用的浓度为50-100 μg/mL。
