    STAT3报告基因是翊圣生物自主研发的用于检测STAT3二聚体和STAT1/STAT3异质二聚体的转录活性水平为目的的报告基因。激活STAT3后形成STAT3二聚体和STAT1/STAT3异质二聚体,它们与SIS-inducible elementSIE)反应元件进行相互作用调控细胞的生长和凋亡。STAT3(signal transducers and activators of transduction-3)报告基因与多种恶性肿瘤的发生、发展和侵袭转移关系密切。

    STAT3报告基因主要用于监控细胞STAT3信号通路中的STAT3转录活性、肿瘤治疗药物的研发以及基因过表达和RNAi的表型分析等。

    pGMSTAT3-GFP是翊圣生物改造后的哺乳动物真核表达载体,在其多克隆位点插入了多个STAT3结合位点,可以高效地检测STAT3的激活水平。由于载体采用了GFP作为报告基因,更便于后续的检测。同时,对载体中预测出的其它转录因子以外的结合位点进行了适当的突变,增加了质粒的转录因子结合特异性。另外,由于质粒体积减小,使得STAT3-GFP报告基因更易于转染。
